Neocortical organoid method and developmental progressions. (A) Neocortical organoid culture method. (B) Area measurement of W2.5 organoids comparing the 6-day ROCK inhibitor Y-27632, 1-day <t>CEPT,</t> and 6-day CEPT treatments. Scale bar: 500 µm. (C-F’) Dynamic laminar layer formation of W2.5 (C-D’) and W5 organoids (E, E’). The basement membrane is marked by LAMININ, and the apical membrane is marked by aPKC. Left panels: whole organoids. Scale bars: 200 µm. Right panels: enlarged images from the boxed region. Scale bars: 50 µm. (F) Astrocyte formation in W12 neocortical organoids, labeled with HEPACAM and GFAP. Scale bar: 50 µm. (F’) Zoom-in images from (F), white dotted square. Scale bar: 20 µm.

Article Snippet: To evaluate the effects of the CEPT small-molecule cocktail , we treated organoids with a combination of 50 nM Chroman 1 (MedChem Express, HY-15392), 5 μM Emricasan (Selleckchem, S7775), polyamine supplement (Sigma-Aldrich, P8483) diluted 1:1,000 following the manufacturer’s instructions, and 0.7 μM trans-ISRIB (Tocris, 5284).
